The fastest and easiest way is to fly; Northwest/Delta will get you from O'Hare to Duluth for about $250.

If you want to do it cheaply, take the Megabus to downtown Minneapolis (see Taking the bus), then Greyhound to Duluth. If you take the 6:30 p.m. Greyhound bus to Duluth, it only takes two hours and 40 minutes, not much more than in a car, and cost is $23-$38.
